Is it supported to follow the following guide for VPC to VPC connectivity between regions within AWS? We already have the access server set up with point to site VPN

Technically, that guide is to connect an existing OpenVPN Access Server on your own local business network, to an AWS network using AWS's own VPN termination service. It uses IPSEC with StrongSwan to do this.
If you want VPC to VPC connectivity, you can use either Amazon's own solutions, or you could do a site to site routing solution with an Access Server and a Linux client as VPN gateway using purely OpenVPN based solutions instead of IPSEC/StrongSwan.
